Louisa May Alcott is a female American Novelist born on November 29, 1832 and in Germantown, Pennsylvania (â€Å"Louisa May Alcott†). She had three sisters, a philosopher/teacher for a father, and a loving Mother (â€Å"Louisa May Alcott†). Her family is said to be her primary inspiration for her success in literature and also served as the foundation for her most popular book, â€Å"Little Women†. Based on the biography written by Stern, the stories of Alcott were lifted from â€Å"episodes from her life, her observations, her travels, her experiences, her reading† (xxii). The use of her own personal insights and experiences have made all of the stories as real as it could be during a time when the society is dominated by men and women are expected to become mere supporters. Nonetheless, Alcott has been a witness to her own strength and that of other women, which allowed her to accomplish a significant number of stories and literature that portrayed women as strong personages existing in a male-dominated society. In order to examine how Alcott has presented women as strong characters in a predominantly male society, her stories, such as â€Å"Little Women† and â€Å"Work: A Story of Experience,† are used to show the distinct power and role that women held in relation to the existing power dynamics between men and women. â€Å"Work: A Story of Experience† Women experienced a pivotal role in the industrial era as workers and as channels for the advocacies of the female working class. In the novel â€Å"Work: A Story of Experience,† the main character, Christie Devon, is shown as a women of great character as she tries to find her rightful place in the industrial age. After her several attempts, she ended up as mediator between the female working class and the owners of the businesses where they work in (Alcott 355). Christie has attributed her distinct capacity to serve as an instrument of negotiation to her mother and father, who are both seen of equally great importance (Alcott 355). In this particular work of Alcott, the author maintained a sense of reality in terms of the existing social structure between men and women in the industries and never denied the presence of gaps between the two. However, women are given the chance to voice out their concerns regarding the oppression they felt, which consequently increases their chances of receiving better treatment. The need for women who are strong enough to bring forth the concerns of the female working class is expressed by Alcott, who states that such women were much needed and not always easy to find; for even in democratic America the hand that earns its daily bread must wear some talent, name, or honor as an ornament, before it is very cordially shaken by those who wear white gloves. (355). In the said quote, there are three distinct elements that show the strength and renewed importance provided for females. First, females are given greater chances of participating in different affairs in the society provided that they have the necessary qualifications to do so. With the said qualifications, they are allowed to express their concerns and make contributes to decisions related to matters that directly affect them. The use of qualifications as a form of permit to engage in matters usually dominated by male is also realized by Christie, who mentioned that the attributes she inherited from both her mother and father are enough for her work as a negotiator. Second, the author recognized the existence of women who have the ability to participate but also noted their inherent rarity. The recognition made by author shows that women have gained strength and shared a certain amount of power albeit in rare instances. It is not to say that it is impossible for women to become as strong as Christie but should strive harder in order to gain a distinct position in the society. In fact, the words of the author do not serve as limitations but of recognition of the presence of strong and able women and implies the need to further increase their number. Third, the author also takes note that there are people in the society with who the females need to be at par with. Those with the â€Å"white gloves† can serve as representations of a class that the women should infiltrate through their qualifications. To a certain extent, it can be understood as the domination of males in the different areas of the society. The congregation of males is seen to be one where affiliation is strictly confounded by certain limitations. Nonetheless, the author still maintained the ability of strong females to break the barriers and become affiliated with the male-dominated fields in the society. Aside from the points mentioned above, the novel also represents women who are able to work outside their houses and take on roles that are different from the ones they usually did inside the household. The women portrayed in this particular novel of Alcott are shown to have the ability to work in different careers and are indeed allowed to do so. It is an indication of their ability to break the common stereotype attached to women at that time, which is to remain inside the household and attend to the needs of the family members. Nonetheless, the women are not spared from their household chores and are still expected to fulfill the usual roles attached to being a mother and a wife. Despite the inherent difficulty, the women are still able to cope with the need to balance both the demands of their work and families that renders them capable of succeeding at different tasks at one time. â€Å"Little Women† The novel, â€Å"Little Women† was published in 1868 and revolved around the story of four different women who are considered to be taken from the experiences of Alcott with her three other sisters (Richards 123). The novel’s main characters, being women, are used by the author to send â€Å"a feminist message about the shared challenges all women face in male-dominated society† (Richards 123). The author placed emphasis on the different barriers that often limited the opportunities made available to women. The obstacles enumerated in the novel are the burden of finding a husband who can serve the needs of the female, restrictions with regard to the choices in terms of career, and problems with regard to access to college education. All four women are shown in the novel to have their own distinct characters and their own means of coping with the difficulties they face as women. Jo, whose experiences are often related to that of the Louisa Alcott, is known to be a tough and free-spirited character (Walker and Dresner 115). All of the other characters are known to be struggling with the factors that limit their capacities. One well-known example is that of Jo who took great leaps in order to fulfill her goals and dreams. The three other women have also shown their own strength as they conquer and fulfill the problems and the expectations. All of them are shown as individuals who are able to work their way out of the comforts of the households and successfully overcame the different dilemmas they faced. The portrayal of the lives and struggles of the four women is successful and also appealed to the senses of the audience. As a result, a sequel to the novel was published and showed a different phase in the lives of the four women. In the second volume, the women are also shown to have their own individual struggles and the means they used to overcome the said struggles. In both volumes of â€Å"Little Women,† the females are presented together with their experiences with regard to the struggles they have to face in a male-dominated society. Interestingly, all of the women are able to face their problems with utmost strength, which shows that women have the capability to overcome the challenges because they have the skills to cope with the said difficulties. Likewise, it allows women to find their strength with other women and to provide the needed comfort and help, especially during difficult times. In fact, there is no other substitute for the kind of care provided by women and the courage they exhibit during critical times. At the time when Beth became ill from scarlet fever, it is her sisters who decided to take care of her. All of them remained strong despite the condition of Beth and took the initiative to solve their problems without needing much help from other people. On another note, Alcott have also shown that despite the dominance of males in the society, the females are still able to succeed in their own professions through their own little ways. In this respect, women are able to show that it is not the number that counts but the skills, knowledge, and abilities that they have. Other Stories In the other stories written by Alcott, the focus is also on the females and how they tried to overcome the problems they face in their own respective fields of profession. An example of this is how a woman is used as the main character for the â€Å"Candy Country. † In the said story, the woman is shown as a person who is capable of acquiring knowledge for the sake of enhancing their own welfare where the author states that â€Å"she goes home and grows up from a ‘sickly, fretful child’ to a ‘fine, strong woman’†¦Ã¢â‚¬  (Alcott xxxv). Despite the simplicity of the story, Alcott ensured that the main character portrays the strength of women despite the domination of men in the society. Conclusion The stories of Alcott are used to represent the strength of women despite the dominance of male in the society. Stories, such as â€Å"Work: A Story of Expereince† and â€Å"Little Women,† the author is able to show the difficulties faced by women and the distinct coping mechanisms carried out in order to solve their problems. Aside from this, Alcott have also shown that women are able to enter aspects of society that are known to be dominated by male. How effectively does Parker translate Act 3 Scene 3 on to the Screen? Essay

This is the first time I have read a Shakespeare play, however I have seen the film version of Romeo and Juliet. At first it was hard to understand, the language was difficult but it was a good story. Parker made the film to suit regular filmgoers, and I found it easier to understand. The plot was gripping gut the language was challenging. The costume, the language and the photography all made it a good Shakespeare experience. As the plot of Othello is more relevant to today than Shakespeare’s time, it includes all the elements of modern day films, and is well presented by Parker. I choose to assess how effectively Parker translates the original script of Act 3 Scene 3, the temptation scene. This is the pivotal scene in the play. At the beginning Othello declares his love for Desdemona, but by the end his mind is set on killing her. This scene also shows the power of Iago and the ability to manipulate all the others. The ‘temptation scene’ is the longest and most important scene in the play. Iago, whose ingenuity, inventiveness, cunning, lack and hypocrisy are evident throughout, plays the dominant role in this scene. An analysis of the various crucial stages in Iagos assault on Othello’s peace of mind, and on the reputations of Desdemona and Cassio, will reveal the depth of Iagos evil genius. At the beginning of the scene Othello is happily married by the end he has decided to murder his wife and Cassio. Iago immediately sets to work, using the slender evidence so far to hand against Cassio. He concentrates first on Cassio’s role as a go-between in Othello’s wooing of Desdemona, and allows Othello to recall the circumstances of Cassio’s departure from Desdemona, and his own reservations about it. Othello has not been favourably impressed by the furtive manner of Cassio’s departure, and is not helped by Desdemona’s insistent, repetitions, irritating pleading on Cassio’s behalf. Iago’s main strategy at this point is to throw out dark, mysterious hints of something that he pretends he does not want to bother Othello with. Iago succeeds in giving the impression that there are some disreputable truths behind his insinuations. Othello’s suspicions inevitably grow, as Iago warns him of the dangers of jealousy. Iago tells Othello that Venetian women are notorious for infidelity to their husbands. He reminds Othello that Desdemona’s deception of her father in marrying him, and points the unnaturalness of her choice of Othello in preference to someone of her own race. Othello is now convinced of Iago’s honesty and knowledge of human nature that he searches for reasons for Desdemona’s infidelity. The sight of Desdemona only causes confusion in Othello’s mind. She notices a distressing change in his attitude and behaviour, and, in trying to help him, drops her handkerchief, a gift from Othello, and a token to which he attaches enormous significance. Emila picks up the handkerchief and gives it to her husband. Iago decides to leave the handkerchief at Cassio’s lodgings. This is going to be Othello’s first sign of truth. Iago, of course, cannot offer any proof, but argues that strong circumstantial evidence ought to be enough to satisfy Othello. He mentions to pieces of manufactured testimony, Cassio’s dream and Cassio’s possession of Desdemona’s handkerchief. Iago was played by Northern Ireland born actor Kenneth Branagh who is a famous Shakespearian actor and director. Irene Jacobs plays the part of Desdemona, who is a lesser-known Belgian actress. Lawrence Fishbourne is the first Black actor to play Othello on film. There are a number of foreign actors in this film, which gives a European appeal and creates the feeling of Italy or somewhere foreign; there is also a wide mixture of foreign accents. Lawrence Fishbourne covers up his American accent very well. His vast array of colourful costumes, earrings, scars and shaven head all suggest his exotic character that has travelled the world. Iago on the other hand has only one costume throughout. This highlights his role as a servant, but even with all the wealth and riches of Othello and Desdemona, Iago is still able to wield his authority over them in to doing what he wants Act 3 scene 3 is 478 lines long, which is long for Shakespeare. In the text the whole thing takes place in the citadel, while Parker uses all the techniques at his disposal to attract and maintain the viewers attention. Parker uses music, a variety of different lighting and camera angels to set the mood. Parker also uses flash back and dream sequences very effectively and has a vast variety of different locations throughout the scene. The first scene has a sense of elegance and gracefulness with the 2 men going down the stairs to the courtyard and up the stairs to Desdemona. Although in this scene Iago throws his first blow, everything is good-natured; there is a feeling of friendliness and harmony. The next scene is the pole fighting between Iago and Othello. Iago is the one who is put to the ground but everything is good-natured. It shows Othello has power. After the pole fighting the scene changes again to the two men working down a narrow stairway washing their hands. Even at this point Othello’s costume has changed. This highlights his power. The scene then changes to the dark, brown, dull armoury, which is full of guns and gunpowder. The bareness of the place, the guns, the cold bare metal and the ever-increasing absence of daylight adds to the ominous feeling of the scene. The camera changes and focuses a lot on the facial expression throughout this scene. A line is added from Act one into this scene, â€Å"†¦ I will pour my pestilence in his ear†¦Ã¢â‚¬  Iagos plan is to pour poison into Othello’s ear. His is the first time that you can clearly see the doubt in Othello’s face. The music then changes to a very high pitched note, suggests something very ominous is about to happen, adds tension. But lightens again when Othello shows he is not going to give into Iagos insinuations, then deepens again to show Othello’s mood. The next scene is up in the bedroom and is even darker, with only a very dim candle to light the room. Iago is dressing Othello for the banquet while Othello is daydreaming, he has suspicious thoughts. While Iago remains the servant, he still exercises power over Othello’s thoughts and feelings. As the scene gets even dimmer Othello begins to succumb to Iagos suggestions. The next scene is the handkerchief scene, which is done in silence. Desdemona and Emila come in and find Othello sitting at the end of the bed sweating. Desdemona wipes his head with the handkerchief and accidentally drops it. When the two leave Emila lifts it. Parker use silence as a powerful symbol in the handkerchief scenes. The handkerchief that seems such an insignificant article proves to be very dangerous in the end. The scene then changes to a bedroom scene with Iago and Emilia, which in contrast to other love scenes in the play, is very crude and vulgar. The room is very dark and small and has little furniture compared to Othello’s bedroom. This emphasizes their role as servants. As Emilia enters Iago turns his head away from her, but when she says she has the handkerchief that Othello gave Desdemona he is all over her, suggesting complete indifference and that he only wants her when she has something for her. At the end of the scene Iago says, â€Å"trifles light as air† and throws the handkerchief up into the air. The next time we see the handkerchief is in Cassio’s lodging where he leaves it in Bianca’s hand before he leaves. Iago had planted it there and Cassio had given it onto Bianca as a gift. Parker then brings a scene in from act four where Othello quizzes Desdemona about the whereabouts of the handkerchief; this is to give more evidence. Othello is sure Desdemona is guilty and then when Desdemona ask Othello to give Cassio his job back this only enrages him further. The scene then changes down to the beach. Parker makes the audience very aware that they are on an island and isolated. The ruggedness of the rocks and the waves and the pounding, shows that they are far from Venice. Iago mocks his master, â€Å"†¦nor poppy nor mandragora†¦Ã¢â‚¬  nothing will bring him back his piece of mind. The climax in his scene is the very dramatic attempted drowning of Iago, also the story where Iago complains of his sore tooth has a very damning affect on Othello. This scene does not appear in the original play by Shakespeare but is very effectively done by Parker to highlight the fact that Othello has totally fallen for Iago’s lies. The next scene is up on the ramparts of the citadel; the feeling of stormy weather emphasizes the mix feelings in Othello’s mind about Desdemona’s distrust. Othello then gets down on both knees and makes a vow to the heavens, very dramatic, tension building. He then says, â€Å"Arise black vengeance, from the hollow hell.† Iago then gets down on his knees and they both swear brotherhood to each other in order to kill two people. Parker adds the slitting of the palms and the clasping of the hands, this increases the horror of what they are going to do. Iago then says, â€Å"I am your own forever.† There is a feel of dramatic Irony, where the audience is aware what is about to happen to Othello but the character does not. The main techniques used by Parker are flash back and dream sequences, which allow us to see inside the mind of Othello, whereas Shakespeare used soliloquy. The flash back and dream scenes are done in silence and don’t last very long. They are done in slow motion and are accompanied by music. They are frequently of Desdemona and Cassio in bed or talking to each other. The lovemaking is done very discreetly and delicacy and are done behind a muslin curtain. They are hinted at rather than stated explicitly. There is a hint of red on the bed, which makes Othello think she is a whore. Music is used effectively by Parker to create an ominous atmosphere and highlights a climax or turning point in the scene. The first time music was introduced was when Othello said, â€Å"It were not for your quiet nor high pitched string note.† The music then deepens to a bass note where Iago says, â€Å"Beware my lord of jealousy† and continues on this deep tone until Othello asks, â€Å"Thinkst thou make a life of jealousy.† The music then becomes more melodious and sweet, suggestion of will not fall prey to Iagos insinuations. At the line, â€Å"Get me some poison† the music deepens again. Parker leaves out about half of the 478 lines and yet he doesn’t compromise the meaning of the play. Very many of Desdemona’s lines are omitted, more than anyone else, and yet with the use of flash back and dream sequences we are very aware of her presence throughout the play. Some of the scenes are rearranged, scene 4 into 3, make Othello’s death sentencing more convincing. Parker does not compromise power and integrity of the play, he delivers a different perspective, which is more creative. There are difficulties with the language but with continued reading I found it a gripping story, and I found Parkers version accessible to modern filmgoers. Shakespeare’s play was limited, the language was used to keep the audiences attention but today film makers have different lighting, music, props and many other things, so this allowed parker to leave much of the language out.

Drosophila Immunohistochemistry Practical Abstract Immunohistochemistry uses monoclonal antibodies to detect specific proteins in tissue samples with a visible label. For this practical, immunohistochemistry was used to visualise the distribution of protein in the 3rd instar Drosophila melanogaster larvae brain. The distribution of protein was visualised using a primary monoclonal antibody, and this primary antibody was then detected by a secondary antibody. Detection of the secondary antibody is via an enzyme reaction which produces a coloured precipitate. The secondary antibody used is a monoclonal antibody that recognises IgG from a mouse which is attached to alkaline phosphatase. Looking at the protein distribution pattern in the larval brain the antibody used was detected. Brain samples were fixed in 4% paraformaldehyde in phosphate buffered saline (PBS) at room temperature for 20 minutes, then washed four times with PBST prior to blocking with 10% donkey serum for 45 minutes. The primary antibody was added, and the brains incub ated at 4 °C overnight. The antibody used was embryonic lethal abnormal vision (ELAV). This is because there are domains in the optic lobe which look like dots and webs from binding of ELAV to the specific proteins in the brain. Introduction Immunohistochemistry is the use of monoclonal and polyclonal antibodies to detect specific proteins in sections of tissues. Applications of immunohistochemistry has been used by pathologists for the diagnosis of cancers. Secondary antibodies are the antibodies which bind to the primary antibody to allow detection of the primary antibody under a microscope (Rockland-inc.com, n.d.). Primary and secondary antibodies are used because primary antibodies detect a complimentary protein which is of particular interest whilst the secondary antibody is raised against immunoglobulins associated with the primary antibody. The secondary antibody then associates with a linker molecule which then recruits reporter molecules or the antibody binds directly to the reporter molecule. Drosophila is used as a model organism because their genome is very closely related to the human genome. This means that human genes can be mapped to drosophila genes. Genes in human s which cause disease are also in drosophila. Many generations can be observed in drosophila because they have a short reproductive cycle. They are small so a lot of them can be kept in a laboratory. Fruit fly genes can be altered very easily. Transgenics can be easily produced in the flies and studied. In tissue preparation, tissues samples are preserved so protein and tissue architecture does not breakdown. Samples may be also need to be perfused for prevention of detecting non-specific proteins which can interfere with the detection of the target protein. In order for protein and tissue to not breakdown fixation is required. Steps of fixation include perfusion, immersion and freezing. In perfusion, tissues are perfused with a fixative to allow rapid fixation. During immersion tissues are immersed in the fixative to allow the fixative solution to diffuse through the tissue or cell sample. In the final step samples with protein that cannot survive fixation are frozen in liquid nitrogen whilst being embedded in a cryoprotective embedding medium. Once fixation is done the sample needs to be sectioned and mounted. The tissues which have been fixed with a paraformaldehyde solution are sectioned into slices which measure 4-5 µm using a microtome. After sectioning the sections are mounted onto a glass slide which is coated with an adhesive. Once mounting has been done the sections are dried and then deparaffinization takes place (Thermofisher.com, n.d.). The next major step is for the target protein to be retrieved. Paraffin is removed in order for the antibodies to reach the protein. Heat induced epitope retrieval is the most common used method to retrieve these proteins. This involves heating the slide at PH6 or PH9 depending on the antibody (Biosciences, 2014). Endogenous blocking needs to place because reagents which are present during immunohistochemistry are also present in cells. This can interfere with protein detection because of unwanted signals being produced that mistakenly detect an protein. Finally, the antibodies are labelled to detect the target antigens. The primary and secondary antibodies are diluted into a buffe r in order for the antibody to be stabilised. Rinse buffers are used between antibody applications to remove unbound antibodies (Thermofisher.com, n.d.). Once they are labelled the samples are viewed under a microscope (Biosciences, 2014). If an nurse that is convicted of one of these crimes may voluntarily surrender there license. When the court requirements are met the convicted nurse may submit a request for a wavier. With the waiver request the nurse must provide evidence that thee court order has been met, rehabilitation, and provide letters of reference.

The organization’s core values are intensely held and widely shared by the whole organization, meaning that all employees believe in and stick to the organizational culture which makes sense why it’s regarded as having a strong culture. Which is reflected through their performance, loyalty and commitment, even when others competitors rose in the market, employees still never left the organization although they demanded better pay. Other reasons might be: Selection: workers were selected on the basis of having a sense of humour and other qualities but sense of humour has been a basic criterion. Top management: always motivated employees by portraying the airline as the underdog. Also allowing a large portion of the compensation to be in the form of stocks ,therefore sharing profits. Finally the provision of job security. Socialization: as mentioned in the case study that the company encouraged it’s employees too work hard and at the same time have a good time. Question two: Do you think upstart airlines can successfully duplicate this culture? The organization should motivate employees to work as hard as they were before. The case now is that part of their compensation is cut due to the decrease of their shares’ value and dividends. And if we are going to look at it, everything happened in a blast but, much unnoticed. In four years of staying here in this institution, I may conclude that counting the wonderful moments that we shared in our high school days is a mind-boggling thing because even the depressing ones that we lived through contributed in helping us to become what we are today.Yes, even being â€Å"pasaways† in front of our teachers, all are considered factors that made this experience complete and of course, a perfect one. And if we are going to look at it, everything happened in a blast but, much unnoticed. When I made my first footsteps in this school in the early days of my freshman year, one thing I can’t forget is our innocent selves, just like a blind person who seeks the right path, looking for the white light behind the darkness of that innoce nce.By all means, we looked for the knowledge we need and we craved for the wisdom we must possess before we depart from this home, from this family. We were so curious about all the things that we encountered everyday but we did not know to whom we must hold on and how to distinguish right from wrong. And because of such blindness, receiving only the radiance of the twilight, some succeeded in knowing all the essential things to be remembered and to be understood by the heart but others got lost and failed to know the indispensable truth of life.We have encountered many things as high school students but still, our journey will not stop after this stage. And I’m very proud that we never lose hope when failures and disappointments come our way. We defended what we love, and fought for what we believed in. And as for you, the juniors, in a few more days, your job will be to continue the unfinished tasks we will leave behind as we sail in our respective ships to discover new oc eans. In behalf of the seniors, we are thankful because you are our companions and these years will serve as memories that we will cherish forever.After we move on, after we step on another phase of our lives, you must learn and remember the things that have helped us to finish this stage. Some things that we did, might not have worked out well but we trust that after we leave the portals of this institution, you may learn from our mistakes. Expect the worst but always strive for the best. Whenever you fail, don’t lose hope, just stand up. Consider such challenges as a blessing. We must learn from these challenges, particularly the failures so that we will achieve more success. You must also learn how to be humble.Humility is one sign of being educated, of being wise. This will be the start of your time to grow up and make your own; we believe that you can do more than what we have done. You must learn to be contented sometimes but grow most of the time. It is always right to be contented but this will sometimes hinder you from growing, from progressing. You only live once, so live your life to the fullest. It is not yet over so never make this as an excuse for you to be contented; there are many more fishes in the sea, waiting for you to discover them.Just be grateful for all your achievements and be thankful to the One who is the first and the last thing we must think of, God, because His love for us and His will to guide us is everlasting. To all Juniors, Knowledge has taught us the elements of character that facilitate success. With this key of acceptance we pass on to you the skills to learn, the aptitude to succeed, and the creativity to make a difference. We hope to see all your spectacular dreams come true.
